RAVE is an international Phase II/III double-blind, placebo-controlled trial of the anti-CD20 monoclonal antibody rituximab in ANCA-associated vasculitis (AAV), specifically Wegener’s Granulomatosis or Microscopic Polyangiitis. The trial is being conducted by the Immune Tolerance Network (ITN), with support from the National Institutes of Health, the Food and Drug Administration and Genentech, Inc.
RAVE aims to assess the efficacy of the monoclonal antibody rituximab to induce remission in patients with ANCA-associated vasculitis (AAV). It will compare the efficacy and safety of rituximab with the current standard of care, cyclophosphamide, when they are used in combination with glucocorticoids.
Preliminary open-labeled studies have shown that rituximab can induce stable remission of AAV.
RAVE will also try to gain a better understand the genetic, cellular and immunologic conditions under which B-cell tolerance to ANCA antigens may be restored. A wide-ranging set of bioassay studies will be carried out on clinical specimens obtained during the trial that will attempt to provide more information on the pathogenesis of the disease.
